Nucleic Acid Reagents RNA Synthesis and Transcription

TriLink's patented CleanCap® Reagent AG (3' OMe) is a co-transcriptional capping reagent for in vitro transcription of 5' capped mRNA resulting in a Cap 1 structure with >95% capping efficiency.

CleanCap® Reagent AG (3′ OMe) is designed for the co-transcriptional capping of messenger RNAs (mRNAs) to produce an optimal Cap 1. CleanCap Reagent AG (3′ OMe) includes the 5′ N7-methyl-3′-O-methylguanosine commonly found in mRNA capped using anti-reverse cap analog (ARCA), whereas CleanCap Reagent AG has a naturally occurring 5′ N7-methylguanosine structure. Using a firefly luciferase mRNA, CleanCap Reagent AG (3′ OMe) shows higher, more widely distributed, and longer sustained in vitro expression than CleanCap Reagent AG.


CleanCap Reagent AG (3' OMe) includes the 5' N7-Methyl-3'-O-Methylguanosine commonly found in mRNA capped using ARCA, whereas CleanCap Reagent AG has a naturally occurring 5' N7-Methylguanosine structure.


Cap-1 mRNAs produced by CleanCap AG (3′ OMe) have superior in vivo activity compared to Cap-0 mRNAs produced by legacy co-transcriptional capping methods such as ARCA. In addition, compared to enzymatic capping, capping with CleanCap AG (3′ OMe) analog offers more streamlined and efficient manufacturing of mRNAs.


CleanCap AG (3′ OMe) can be used in conjunction with TriLink’s modified and unmodified NTPs, as well as in vitro transcription (IVT) enzymes. Using the recommended conditions, transcription with CleanCap AG (3′ OMe) results in >95% capped material, a Cap-1 structure, and crude yields of 4 to 5 mg/ml of transcription.


CleanCap Reagent AG (3’ OMe) require an AG initiator.
