Supplier: Biotium
Description: This antibody recognizes human blood group A (monofucosyl and difucosyl A antigens with chain types 1, 2, 3, 4, 5, 6) and Forssmann antigen. It is also reactive with the immuno-dominant A trisaccharide. Blood group antigen expression in human colon cancer was studied by means of two monoclonal antibodies of broad anti-A (HE-14) and anti-type 3 and type 4 chain-based A and H (HE-10) specificity. These antigens were proved to re-appear in tumors of the distal colon, the HE-10 antibody reacting more frequently (9 out of 12 samples) than HE-14 (5 out of 12 samples) and frequently with supra-nuclear staining of the cytoplasm probably in those places of the Golgi apparatus where carbohydrate antigens are synthesized. This staining pattern is characteristic of HE-10 in normal colonic mucosa as well. With HE-14, staining was often absent in less differentiated tumors, while HE-10 did react in such tumors. In some cases, these two antibodies gave different staining patterns in parallel sections from the same tissue sample, primarily at the cellular level. Three out of 12 cases showed blood group antigen expression in the mucosa of the distal colon adjacent to the tumor only when HE-10 Ab was used.

Description: The claudin superfamily consists of many structurally related proteins that are important structural and functional components of tight junctions. Claudin-12, also known as CLDN12, is a 244 amino acid multi-pass membrane protein that is expressed in the brain, duodenum, jejunum, ileum, and colon. Claudin-12 localizes to cell junctions and may be involved in tight junction integrity by regulating hyperammonemia. Ammonia can alter brain capillary endothelial cell gene expression and transporter function. Claudin-12 is upregulated in enterocytes through vitamin D receptors which strongly suggest that claudin-12 forms paracellular Ca(2+) channels in intestinal epithelia and may be critical for vitamin D-dependent calcium homeostasis.

Description: Exhibits intrisinic GTPase activity. Shows a higher affinity for GDP over GTP (about 12-fold higher), and binding shows an absolute requirement for magnesium.Tissue specificity:Highly expressed in spleen and peripheral blood leukocytes that contain mostly T- and B-lymphocytes. Expressed specifically in resting T- and B-lymphocytes and expression significantly decreases during B- or T-lymphocyte activation. Expressed at lower levels in thymus, ovary, colon and small intestine.

Description: DDAH, a dimethylarginine dimethylaminohydrolase, hydrolyzes dimethyl arginine (ADMA) and monomethyl arginine (MMA), both inhibitors of nitric oxide synthases, and may be involved in in-vivo modulation of nitric oxide production (1,2). Impairment of DDAH causes ADMA accumulation and a reduction in cGMP generation (3). DDAH II, the predominant DDAH isoform in endothelial cells, facilitates the induction of nitric oxide synthesis by all-trans-Retinoic acid (atRA) (4). DDAH proteins are highly expressed in colon, kidney, stomach and liver tissues (1).

Description: SN-38 is the active metabolite of irinotecan, a derivative of the alkaloid camptothecin and acts as a potent inhibitor of DNA topoisomerase I. SN-38 is a more potent topopisomerase I inhibitor and more cytotoxic to HT-29 colon cancer cells (IC50 = 8.8 nM) compared to irinotecan (IC50 >100 nM).

Description: CAPS2 is a 557 amino acid calcium-binding protein that is abundantly expressed, with highest expression found in placenta, testis, colon, lung and brain. CAPS2 contains three EF-hand domains and exists as three alternatively spliced isoforms. Suggested to play a role in large dense-core vesicle (LDCV) exocytosis, CAPS2 is encoded by a gene that maps to human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome. Chromosome 12 is associated with a variety of diseases and afflictions, including hypochondrogenesis, achondrogenesis, Kniest dysplasia, Noonan syndrome and trisomy 12p, which causes facial developmental defects and seizure disorders.
