Description: 2,2-Azino-bis-(3-ethyl-benzthiazoline-6-sulfonic acid) (ABTS) has been used as a chromogenic substrate for horseradish peroxidase (HRP), both in general activity assays and in ELISA applications. Activity of HRP using ABTS appears about four-fold higher than using pyrogallol. It is mainly used as a substrate in sensitive peroxidase assays.
2,2′-Azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) is a peroxidase substrate suitable for use in ELISA procedures. This substrate produces a soluble end product that is green in color and can be read spectrophotometrically at 405 nm. The reaction may be stopped with 1% sodium dodecyl sulfate (SDS). Recommended for ELISA (microwell) procedures, not recommended for membrane applications.

D-Biotin is a growth factor present in small amounts in every living cell. It is involved in naturally occurring carboxylation reactions. It occurs mainly bound to proteins or polypeptides. It is more abundant in the liver, kidney, pancreas, yeast and milk. Biotin levels are higher in cancerous tumors than in normal tissues. It is inactivated by binding to avidin.
D-Biotin may be used to elute proteins from avidin/streptavidin resins. It is widely used for dietary supplements and fortified foods. It is also used for tablets and hard-shell capsule preparation due to its pharmaceutical properties.

Description: SMPT is a heterobifunctional sulfhydryl- and amine-reactive cross-linker. SMPT is used often for conjugating a toxin molecule to a monoclonal antibody directed against a cell-surface antigen. These immunotoxins produced with SMPT are highly potent as the cleavable disulfide imparts increased cytotoxicity compared to conjugates without a cleavable bond, such as those produced with SPDP. Also, next to the pyridine-2-thione in the spacer arm are a benzene ring and a methyl group adjacent to a carbon that hinders the disulfide bond, allowing exceptional conjugate stability in vivo. SMPT has the added benefit of being more stable to hydrolysis in aqueous solutions than other NHS cross-linkers.

Description: The amine-reactive portion of SPDP reagents is the N-hydroxysuccinimide (NHS) ester. Reactions are most commonly performed in phosphate, carbonate/bicarbonate, or borate buffers at pH 7-8. Other buffers can be used provided they do not contain primary amines (or thiols or disulfide reducing reagents – see discussion of the sulfhydryl reactivity). The rate of reaction and degradation by hydrolysis increases with increasing pH; for example, the half-life of the NHS ester is several hours at pH 7 and less than 10 minutes at pH 9. NHS-ester reagents like SPDP and LC-SPDP have limited aqueous solubility and must be dissolved in organic solvent before adding them to a reaction mixture. Sulfo-NHS-ester reagents like Sulfo-LC-SPDP are water-soluble and can be added directly to aqueous reaction mixtures.
