Description: Tyrosine-protein kinase that acts as cell-surface receptor for the cytokine KITLG/SCF and plays an essential role in the regulation of cell survival and proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration and function, and in melanogenesis. In response to KITLG/SCF binding, KIT can activate several signaling pathways. Phosphorylates PIK3R1, PLCG1, SH2B2/APS and CBL. Activates the AKT1 signaling pathway by phosphorylation of PIK3R1, the regulatory subunit of phosphatidylinositol 3-kinase. Activated KIT also transmits signals via GRB2 and activation of RAS, RAF1 and the MAP kinases MAPK1/ERK2 and/or MAPK3/ERK1. Promotes activation of STAT family members STAT1, STAT3, STAT5A and STAT5B. Activation of PLCG1 leads to the production of the cellular signaling molecules diacylglycerol and inositol 1,4,5-trisphosphate. KIT signaling is modulated by protein phosphatases, and by rapid internalization and degradation of the receptor. Activated KIT promotes phosphorylation of the protein phosphatases PTPN6/SHP-1 and PTPRU, and of the transcription factors STAT1, STAT3, STAT5A and STAT5B. Promotes phosphorylation of PIK3R1, CBL, CRK (isoform Crk-II), LYN, MAPK1/ERK2 and/or MAPK3/ERK1, PLCG1, SRC and SHC1.

Description: SIRP alpha Antibody: Protein tyrosine phosphatases (PTPases) SHP-1 and SHP-2 are critical regulators in the intracellular signaling pathways that result in cell responses such as mitosis, differentiation, migration, survival, transformation or death. SHP-2 is a signal transducer for several receptor tyrosine kinases and cytokine receptors. A novel SHP-2 associated glycoprotein was recently cloned from human, rat, mouse and cattle by several labs and was designated SIRPalpha, SHPS-1, MyD-1, BIT and p84. SIRPalpha is a new gene family containing at least fifteen members. SIRPalpha is a substrate of many activated tyrosine kinases such as insulin receptor, EGFR, PDGFR and src, and a specific docking protein for SHP-2. SIRPalpha has regulatory effects on cellular responses induced by serum, growth factors, insulin, oncogenes, growth hormones and cell adhesion and plays a general role in different physiological and pathological processes.

Description: B-Cell Differentiation Antigen CD72 (CD72) is a single-pass type II membrane protein. CD72 exists as a disulfide-linked homodimer and contains one C-type lectin domain. CD72 is expressed on B lineage cells, NK cells, monocytes, dendritic cells, and mast cells. CD72 is a ligand for CD5. CD72 associates with CD5, interacts with PTPN6/SHP-1 and plays a role in B-cell proliferation and differentiation. CD72 associates with CD79A in the B cell antigen receptor (BCR) complex following antigen stimulation and dampens BCR signaling through interactions with the phosphatase SHP-1.

Description: Siglec11 Antibody: Siglecs are sialic acid-binding lectins of the immunoglobulin superfamily that are mainly expressed in cells of the hematopoietic system. Siglec11 is unlike other siglecs in that it binds specifically to alpha2-8-linked sialic acids and is not found in peripheral blood leukocytes but instead on macrophages of various tissues. Siglec11 is highly homologous to Siglec10 over their extracellular domain but not over their cytoplasmic domain, suggesting that Siglec11 arose through gene duplication followed by a recombination event involving another ancestral siglec gene. Following treatment of Siglec11-transfected cells with pervanadate, Siglec11 becomes tyrosine-phosphorylated and strongly associates with SHP-1 and SHP-2. At least five isoforms of Siglec11 are known to exist. Siglec11 antibody will not cross-react with Siglec10.

Description: LMIR1, also termed CD300a, is a type I transmembrane glycoprotein with a single IgV-like extracellular domain and an extended membrane proximal region that links the immunoglobulin (Ig) and transmembrane domains and belongs to the immunoglobulin superfamily. The intracellular domain of LMIR1 contains several immunoreceptor tyrosine-based inhibition motifs (ITIMs). When cross-linked, it will be tyrosine phosphorylated and capable of recruiting tyrosine phosphatases (SHP-1, SHP-2) and inositol polyphosphate 5-phosphatase, SHIP. LMIR1 will regulate mast cell-mediated inflammatory responses. LMIR1 is broadly expressed on myeloid and lymphoid cells, and its expression is differentially regulated depending on the cell type.
