Description: Vesicle-associated membrane protein (VAMP), also known as synaptobrevin, is an 18 kDa integral membrane protein localized to the cytoplasmic surface of synaptic vesicles. Two VAMP homologs, VAMP1 and VAMP2, are differentially expressed in the nervous system. VAMP2 is found in neurons associated with autonomic, sensory and integrative functions, and in non-neuronal Langerhans islets and glomerular cells. VAMP is a component of a 20S complex implicated in vesicle docking and fusion, the formation of which involves the interaction of VAMP with syntaxin through an ATP-dependent reaction modulated by synaptotagmin, SNAP-25, alpha-SNAP, and NSF.

Description: rSec6 and rSec8 are rat homologs of the yeast Sec6 and Sec8 proteins that are essential for protein transport between the Golgi apparatus and the plasma membrane. In yeast, Sec8 is part of a 19.5S complex that includes Sec6 and Sec15, that also associate with the ras-like GTP binding protein Sec4. An analogous 17S complex in rat brain was found to contain rSec6 and rSec8, along with eight proteins with a combined molecular weight of 743 kDa. The membrane-associated rSec6/8 complex co-immunoprecipitates with syntaxin, providing evidence that this complex is involved in synaptic vesicle docking and fusion at the presynaptic terminal.

Description: Osteopontin (OPN) is an acidic extracellular matrix cell adhesion protein that is relatively abundant not only in bone matrix, plasma, urine, and milk, but is also found in malignant and atherogenic tissues. Phosphorylation, glycosylation and calcium modifications allow intact and fragmented Osteopontin to direct a variety of diverse responses including tissue remodeling, inflammation and cell survival. Plasma Osteopontin has been shown to provide prognostic information in breast, prostate, colon, and lung cancers as well as metastatic carcinomas. The notable presence of Osteopontin in a variety of tumors is strongly correlated to pathological stage, suggesting its critical role in tumor invasiveness, progression and metastasis. In addition, Osteopontin inhibits inducible nitric oxide synthase activity, thereby protecting tumor cells from NO-mediated macrophage cytotoxic attack. Osteopontin is found in atherosclerotic plaques and may drive a number of diabetic vascular pathologies.
