Description: The complement system provides innate defense against microbial infection and is a "complement" to antibody mediated immunity. The complement system consists of thirty five interacting plasma and membrane associated proteins which contribute to host-defense and initiate and amplify inflammation, even in the preimmune state where specific antibodies and lymphocytes are not available. In addition to the complement components themselves, this system also contains several soluble factors that prevent spontaneous complement activation from occurring in solution, as well as, several regulatory membrane associated proteins that protect host cells from accidental complement attack. Cleavage of the C3 component by either the classical pathway or the alternative pathway releases C3a and C3b. C3a appears to be important in many inflammatory responses while the C3b fragment covalently binds to the cell or bacterial surface and plays a role in opsonisation. Binding of C3b to the C4b component of the C3 convertase, results in C5 convertase (C4b3b2a) formation

The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer is standardized to an in-house standard by equivalence-point precipitation. Each IgG fraction is tested for purity and specificity at a minimum of 40 mg/mL using immunoelectrophoresis. The product is mostly goat IgG; no trace of albumin is detected.

This product is suitable for use as a primary reagent in enzyme immunoassays, fluorescein immunoassays and blot, cell or tissue immunostaining. Note: F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors. The product has been successfully used in Immunoblotting. Glycine is a non-essential amino acid. It is only amino acid with no asymmetric carbon and thus is not chiral. It is the major inhibitory neurotransmitter. It is involved in the biosynthesis of the porphyrin rings of hemes and chlorophylls.
Glycine is commonly used in buffer solutions, in electrophoresis and preparative chromatography. A study of the folding of monoclonal antibodies in the presence of glycine and their subsequent purification has been published. The use of glycine in the purification of lipopolysaccharides, lipooligosaccharides, and lipid A has been reported. It is an amino acid for use in cell culture media development applications and existing media formulations. Glycine is commonly used as a component in Tris-glycine and Tris-glycine-SDS running buffers for polyacrylamide gel electrophoresis, a component of Towbin's transfer buffer for Western blots, a buffer substance in cryoenzymology, in osmotic pressure maintenance in isoelectric focusing of erythrocytes, salting-in effect in protein chemistry, and as a buffer component in the coupled phosphatase-kinase reaction for end labelling of restriction fragments. Description: SDS is an anionic detergent and wetting agent that is effective in both acid and alkaline solutions.
Used to solubilize and denature proteins for denaturing-PAGE. Most proteins bind SDS in a ratio of 1.4 g SDS per gram of protein. The charges intrinsic to the protein become insignificant compared to the overall negative charge provided by the bound SDS. The charge to mass ratio is essentially the same for each protein and will migrate in the gel based only on their size.

Description: Elastase is prepared from porcine pancreas. It is lyophilised and water soluble. Elastase is chromatographically prepared by the method of Narayanan and Anwar. In the method, 2 times crystalline elastase is adsorbed on a column of DEAE-Sephadex A50 to separate elastase and non-specific protein components. The elastase component is further purified by chromatography on a column of CM-Cellulose using a sodium chloride gradient to elute the elastase. The latter is dialysed until chloride-free and then lyophilised. During its preparation the elastase is held below a pH of 5,5 for greater than 24 hours. Two times crystallised from the euglobin fraction of porcine pancreas by the method of Lewis et al. Does not contain trypsin or chymotrypsin.
