Description: The Fluoro™ Protease Assay Kit is designed for the quantitative determination of proteases present in a protein sample. The assay uses fluorescein isothiocyanate (FITC)-labelled casein as a general protease substrate. The fluorescein label on the FITC-casein is highly quenched. When the proteases present in the sample of interest digest the FITC-casein substrate into smaller peptides, the quenching of the fluorescence label is relieved and the fluorescence of the substrate is increased. The fluorescence of the FITC-labeled peptide is measured with excitation at 485 nm and emission at 530 nm to determine protease activity. The kit detects picogram level of proteases present in the sample.

Description: Lipid peroxidation is a well-established mechanism of cellular injury in both plants and animals, and is used as an indicator of oxidative stress in cells and tissues. Lipid peroxides, derived from polyunsaturated fatty acids, are unstable and decompose to form a complex series of compounds. These include reactive aldehydes, of which the most abundant is malondialdehyde (MDA). Therefore, measurement of malondialdehyde is widely used as an indicator of lipid peroxidation. Increased levels of lipid peroxidation products have been associated with a variety of chronic diseases in both humans and model systems.

Description: Glucose Oxidase is a dimeric enzyme that binds to β-D-glucose and aids in its oxidation into D-glucono-1,5-lactone which then hydrolyzes to gluconic acid. Flavin adenine dinucleotide (FAD) is a cofactor to Glucose Oxidase that acts as the initial electron acceptor and is required for this oxidation to occur. Glucose Oxidase is a natural preservative found in honey, where it reduces atmospheric oxygen into hydrogen peroxide which acts as an antibacterial barrier. Glucose Oxidase is also commonly used in biosensors in which it conveys levels of glucose by keeping track of the number of electrons passed through the enzyme. In this application, Glucose Oxidase is connected to an electrode and the resulting charge is measured.

Description: Glutathione (GSH) is a tripeptide antioxidant which reduces disulfide bonds between cytoplasmic proteins. The constitutive enzyme glutathione reductase transforms glutathione into its reduced state which ultimately can provide a measure of cellular toxicity. GSTT2 (glutathione S-transferase theta-2), also known as GST class-theta-2, is a 244 amino acid enzyme with sulfatase activity that functions in conjugating reduced glutathione to hydrophobic electrophiles. GSTT2 exists as a homodimer in the cytoplasm and is expressed in low levels in the liver and the lung. GSTT2 belongs to the GST superfamily and contains both a GST C-terminal and a GST N-terminal domain. The gene encoding GSTT2 exists on human chromosome 22.

Description: The Biosensis ApoE/Aβ Complex ELISA kit is a sandwich ELISA that allows the preferential measurement of ApoE/Aβ complexes. This kit consists of a pre-coated mouse monoclonal anti-Aβ capture antibody, a highly validated ApoE/Aβ complex standard that is pre-formed, lyophilised and ready for reconstitution, a biotinylated ApoE detection antibody, and horseradish peroxidase (HRP)-conjugated streptavidin and detection reagent. The addition of a substrate (3,3’,5,5’-tetramethylbenzidine, TMB) yields a coloured reaction product which is directly proportional to the level of ApoE/Aβ complex present in samples and protein standards. Importantly, a well-characteriaed and unique ApoE/Aβ complex is included as a standard. This complex is pre-formed and lyophilised, requiring only reconstitution with assay diluent prior to use.

Description: Neuronal Marker Doublecortin (DCX) is a microtubule-associated protein expressed almost exclusively in immature neurons. Neuronal precursors begin to express DCX shortly after exiting the cell cycle, and continue to express DCX for 2-3 weeks as the cells mature into neurons. Downregulation of DCX begins after 2 weeks, and occurs at the same time that these cells begin to express, a marker for mature neurons. Due to the nearly exclusive expression of DCX in developing neurons, this protein has been used increasingly as a marker for neurogenesis. Indeed, the levels of DCX expression increase in response to exercise, which occurs in parallel with increased BrdU labelling, currently a "gold standard" in measuring neurogenesis.

Description: Alpha-fetoprotein (AFP) is a major plasma protein produced by the yolk sac and the liver during fetal life. Alpha-fetoprotein expression in adults is often associated with hepatoma or teratoma. However, hereditary persistance of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the alpha-fetoprotein and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. Alpha-fetoprotein is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of alpha-fetoprotein in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly.
